胰腺癌(PC)是全球性的健康問題,生存率非常低。UL16結合蛋白2(ULBP2)是一種新的PC生物標誌物。本研究目的為開發一套簡單、可靠和價格低廉的胰臟癌阻抗檢測系統,主要分成免疫生物感測器以及讀出電路兩個部分。運用絲網印刷技術製作ULBP2免疫生物感測器。串聯排列可以提高免疫生物感測器靈敏度和訊雜比(SNR)。對於1x1陣列免疫生物感測器,它具有低檢測極限(4.7pg / mL)和高靈敏度(18.5Ω/ Log(pg / mL)),線性(R2 = 0.96),選擇性(> 90%)和長期儲存穩定性(儲存28天後保持95%活性)。讀出電路為利用微處理器控制阻抗讀出電路及類比數位轉換器,並將操作步驟及結果顯示於顯示器上,本系統的檢測結果具有與市售阻抗分析儀WK6420C相當的準確性。總而言之,本研究設計出的免疫生物感測器可以準確地測定ULBP2抗原濃度,且讀出電路系統具有良好的準確性,應用於早期胰臟癌的檢測,達到早期發現早期治療的目的。

Pancreatic cancer (PC) is a global health problem and it has a very low survival rate. And, UL16 binding protein 2 (ULBP2) is a new biomarker for PC. We have developed a simple, reliable and inexpensive impedance detection system, which is mainly divided into two parts: immunosensor and readout circuit to detect PC. The ULBP2 immunosensor was fabricated using screen-printed technology. The immunosensor has a sensitivity of 18.5 Ω/Log(pg/mL), with good linearity (R2 = 0.96) and long-term storage stability (retaining 95% activity after 28 days storage). The readout circuit is based on microprocessor to control impedance readout circuit. Evaluations showed that the readout circuit has good accuracy as compared with a commercial impedance analyzer. It is therefore concluded that the designed immunosensor can determine the concentration of ULBP2 antigen accurately and the readout circuit has good accuracy.
